Total Coliform and E-coli

Obj:

1.) Tounderstand & implement method to prove the existing of e-coli

2.) Thequantity of the presence of e-coli & T.c in water samples.

Coliform: Indicate hamfrul microorganisms to human, ifTC found in water, vege, soil contaminated with harmful microorganism.

E-coli:Commonlyfound in GUT of warm-blooded animal

· Sample turns yellow when coliform are present and fluoresces to indicate presence of E-coli.

Apparatus

2. Incubator

3. 365nmUV light –check TC & e-coli

Procedure

1. Poured 100ml w. samples

2. Pack of colilert added

3. Mix and shake

4. Poured into quantity tray

5. Remove bubble by taping

6. Sealed quantity tray

7. Placed into incubator 24hrs

8. Total of e-coli calculated

Presentof e-coli indicates contamination.

Contaminateddrinking water with these can cause stomach illness such Nausea, and lead todeath.

Prevention

· Boil water

· Chlorination, distillation, disinfection

Total Coliform	E-Coli
· Included bacteria that are found in the soil in water that has been influenced by surface water · used to indicate water quality · Most of TC are not considered pathogens under normal conditions	· Major species in fecal coliform group · Generally not found growing and reproducing in the environment · Considered to be species of coliform bacteria that is the best indicator
